I want to add some text "Please do not reply to this email" to my New announcement template. I found it in Themes/Default/Languages/Email.Templates.english.php
I added a line of text but it didn't work.
Can this be done? And could it be highlighted in capitals, underlined or coloured red?

How and where did you add the line of text?

there are several similar versions of email templates in that file, are you sure you modified the correct one?

I've solved this by editing Email.Templates.english_british.php instead of Email.Templates.english.php

Well, sure... if you are using British English,  you have to edit the correct language file

Ah, I didn't realize that. To prevent future confusion, I've made English my default language.
